=== Lymphangioleiomyomatosis === In May 2015, the FDA approved sirolimus to treat lymphangioleiomyomatosis (LAM), a rare, progressive lung disease that primarily affects women of childbearing age. This made sirolimus the first drug approved to treat this disease. LAM involves lung tissue infiltration with smooth muscle-like cells with mutations of the tuberous sclerosis complex gene (TSC2). Loss of TSC2 gene function activates the mTOR signaling pathway, resulting in the release of lymphangiogenic growth factors. Sirolimus blocks this pathway. The safety and efficacy of sirolimus treatment of LAM were investigated in clinical trials that compared sirolimus treatment with a placebo group in 89 patients for 12 months. The patients were observed for 12 months after the treatment had ended. The most commonly reported side effects of sirolimus treatment of LAM were mouth and lip ulcers, diarrhea, abdominal pain, nausea, sore throat, acne, chest pain, leg swelling, upper respiratory tract infection, headache, dizziness, muscle pain and elevated cholesterol. Serious side effects including hypersensitivity and swelling (edema) have been observed in renal transplant patients. While sirolimus was considered for treatment of LAM, it received orphan drug designation status because LAM is a rare condition. The safety of LAM treatment by sirolimus in people younger than 18 years old has not been tested.
==== Public opinion on impeachment ==== In a survey conducted by Economist/YouGov, half of the American public (50%) expressed strong or somewhat support for the impeachment of Defense Secretary Pete Hegseth, citing his decision to initiate military action against Iran without congressional authorization. Roughly three in ten Americans (30%) indicated opposition to such a move. This sentiment, however, is far from uniform across the political spectrum. Democrats demonstrated the highest level of support for impeachment, with 77% in favor and only 14% opposed. Independents also leaned toward impeachment, with 50% in support and 22% against. Republicans, by contrast, largely rejected the idea, opposing impeachment by a margin of 53% to 26%.

== Structure == In humans, the gene for StAR is located on chromosome 8p11.23 and the protein has 285 amino acids. The signal sequence of StAR that targets it to the mitochondria is clipped off in two steps with import into the mitochondria. Phosphorylation at the serine at position 195 increases its activity. The domain of StAR important for promoting cholesterol transfer is the StAR-related transfer domain (START domain). StAR is the prototypic member of the START domain family of proteins and is thus also known as STARD1 for "START domain-containing protein 1". It is hypothesized that the START domain forms a pocket in StAR that binds single cholesterol molecules for delivery to P450scc. The closest homolog to StAR is MLN64 (STARD3). Together they comprise the StarD1/D3 subfamily of START domain-containing proteins.

== Side effects == The main side effects of relugolix for uterine fibroids include abnormal uterine bleeding (24.6–48.6% vs. 6.3% for placebo), hot flashes (42.8–45.5% vs. 0% for placebo), heavy menstrual bleeding (12.1–49.3% vs. 9.4% for placebo), headache (12.3–15.2%), and excessive sweating (9.4–15.2% vs. 0% for placebo). In addition, decreased bone mineral density occurs with relugolix (21.7% decrease by week 12, 24.4% decrease by week 24).
