Tesamorelin is a synthetic peptide that acts as an analog of growth hormone-releasing hormone, a natural hypothalamic signal. Its sequence corresponds to the forty-four amino acid form of the human hormone, with a small acyl group attached near the amino terminus. That modification slows enzymatic breakdown and extends the time the peptide remains active in circulation. The compound was developed as a pharmacological way to raise endogenous growth hormone output rather than supplying the hormone directly.
After injection, the peptide binds receptors on somatotroph cells in the anterior pituitary. Receptor activation raises intracellular cyclic AMP and triggers release of stored growth hormone into the bloodstream. Because the compound works through the body's own regulatory system, growth hormone pulses retain much of their normal feedback control. Repeated administration also raises insulin-like growth factor 1, a hormone produced mainly in the liver. Investigators treat that rise as a marker that the pituitary axis has been engaged.
The hexenoyl cap slows the enzyme step that trims the amino terminus of native GHRH, the same step that shortens its active lifetime in circulation. As a result, the modified peptide persists longer in plasma than the unmodified hormone in side-by-side comparison. Receptor activity stays broadly comparable, because the added group sits away from the residues that contact the binding site. This combination, preserved receptor activity with reduced degradation, explains why the analog was developed instead of the native sequence.
Several compounds share the GHRH framework, including sermorelin, the shorter 1-29 fragment, and other analogs built on the full 1-44 chain. Naming follows a common convention: a stem that identifies the peptide plus a suffix marking analog status. Reports may describe tesamorelin by its sequence fragment, as a GHRH(1-44) analog, or by its amino-terminal modification. Indexing the compound therefore requires searching all of these forms, since some older literature predates the current international nonproprietary name.

A documented effect of tesamorelin is a reduction in visceral adipose tissue in some study populations. Researchers have reported decreases in trunk fat measured by computed tomography alongside changes in lipid markers. The mechanism is thought to involve growth hormone-mediated lipolysis, though the precise contribution of direct versus indirect pathways is not fully resolved. Studies have generally examined defined groups over finite periods, so long-term outcomes are less well characterized. Findings have not been uniform across all trials.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone (GHRH). Its sequence corresponds to the 44-amino-acid form of human GHRH with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification slows enzymatic cleavage and extends the peptide's activity relative to the native hormone. The compound is produced by solid-phase peptide synthesis and supplied as a lyophilized powder. Researchers classify it as a GHRH receptor agonist. Its structure places it in the same family as other growth hormone secretagogues that act on the pituitary.
Binding of tesamorelin to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and the release of growth hormone into circulation. Because the peptide acts upstream of the growth hormone axis, its effects are partly mediated by hepatic insulin-like growth factor 1 (IGF-1) production. The pulsatile character of endogenous growth hormone secretion is preserved rather than replaced. Whether amplified signaling produces effects beyond those of native GHRH remains an area of ongoing investigation.
Tesamorelin occupies a narrow position among agents that act on the growth hormone axis. Unlike growth hormone itself, which is given as replacement, it stimulates the pituitary to release the hormone in pulses, so the downstream increase in insulin-like growth factor 1 depends on intact somatotroph function. Other peptides in the same family include shorter GHRH fragments and synthetic secretagogues with different stability profiles. Several points remain unresolved, including whether the reduction in visceral fat translates into fewer cardiovascular events, what happens to metabolic markers after long-term use, and how the drug compares with lifestyle or surgical approaches.
Tesamorelin is a synthetic peptide that belongs to the growth hormone-releasing hormone family and contains the same forty-four amino acid sequence as endogenous GHRH, extended at the amino terminus by a trans-3-hexenoyl group. That small fatty acid modification protects the peptide from rapid cleavage by dipeptidyl peptidase-4, the enzyme that shortens the half-life of native GHRH to only a few minutes. Chemically the compound is produced by solid-phase peptide synthesis, purified by chromatography, and supplied as a sterile lyophilized powder for reconstitution.
=== Upstanders === In September 2016, Starbucks announced a debut of its first-ever original content series called "Upstanders", which aimed to be inspirational. The series featured podcasts, written word, and video, and was distributed via the Starbucks mobile app, online, and through the company's in-store digital network.
The receptors for the RLN/INSL peptides are collectively called "Relaxin family peptide receptors (RXFPs)". In humans there are four RXFP receptors (RXFP1-4) all of which are cell membrane-associated and coupled to G-proteins (known as G protein-coupled receptors or GPCRs). There are two distinct families of RXFPs: RXFP1 and RXFP2 are evolutionarily related to the receptors of follicle-stimulating hormone (FSH) and LH, and are the cognate receptors for RLN and INSL3 respectively in humans. On the other hand, RXFP3 and RXFP4 are related to somatostatin and, in humans, are the cognate receptors for RLN3 and INSL5. There is evidence that some relaxin hormones may also be able to interact with glucocorticoid-type nuclear receptors, which float freely between the cytoplasm and nucleoplasm.
=== Topical use === The most common adverse events associated with the use of topical tacrolimus ointments, especially if used over a wide area, include a burning or itching sensation on the initial applications, with increased sensitivity to sunlight and heat on the affected areas. Less common are flu-like symptoms, headache, cough, and burning eyes.
Aquino was assassinated at Manila International Airport (now Ninoy Aquino International Airport). September 1: Civilian Korean Air Lines Flight 007, with 269 passengers, including U.S. Congressman Larry McDonald, is shot down by Soviet interceptor aircraft. September 26: The 1983 Soviet nuclear false alarm incident occurs. The U.S.S.R. nuclear early warning system reports launch of multiple U.S. intercontinental ballistic missiles. Stanislav Yevgrafovich Petrov, an officer of the Soviet Air Defence Forces, correctly identifies them as false alarms. This decision is seen as having prevented a retaliatory nuclear attack based on erroneous data on the United States and its NATO allies, which likely would have resulted in nuclear war and the deaths of hundreds of millions of people. October 25: U.S. forces invade the Caribbean island of Grenada in an attempt to overthrow the Communist government, expel Cuban troops, and abort the construction of a Soviet-funded airstrip. November 2: Exercise Able Archer 83 – Soviet anti-aircraft misinterpret a test of NATO's nuclear warfare procedures as a fake cover for an actual NATO attack; in response, Soviet nuclear forces are put on high alert. December 10: The National Reorganization Process military junta of Argentina is dissolved by democratically elected president Raúl Alfonsín.

=== Liquefaction === The liquefaction is the process when the gel formed by proteins from the seminal vesicles and the prostate is broken up and the semen becomes more liquid. It normally takes between 30 minutes and 1 hour for the sample to change from a thick gel into a liquid. In the NICE guidelines, a liquefaction time within 60 minutes is regarded as within normal ranges.
=== Pharmacology applications === The development of NanoSIMS for organo-metallic drugs paved the way for exploring the distribution of biologically active molecules at the subcellular level. Legin et al. combined NanoSIMS with fluorescence confocal laser scanning microscopy to characterize the subcellular distribution of 15N isotopically labeled Pt-bearing cisplatin in human colon cancer cells. Cisplatin appears in the targeted nucleus of the colon cancer cells. 15N and Pt are separated showing subcellular metabolism is in the path of action. The internalization of amiodarone into the lysosomes of macrophages is illustrated in Jiang et al. Thanks to low detection limit, two iodine atoms of 127I in amiodarone molecule enables a label-free imaging by NanoSIMS. Iodine and phosphorus imaging along with plotting the intensity of 127I− vs 31P− indicated a linear relationship between the amount of iodine and phospholipids. These results disclose evidence of amiodarone-induced phospholipidosis. He et al. visualized the distribution of therapeutic antisense oligonucleotides labelled with bromine (Br-ASO) in some varieties of cultured cells and importantly mouse tissues (heart, kidney, and Liver) using NanoSIMS data combined with back scattered electron microscopy. They demonstrated that phosphorothioate ASOs associate with filopodia and the inner nuclear membrane of cells. They also documented essential cellular and subcellular heterogeneity in ASO distribution in the mouse tissues. Becquart et al.
=== Pharmacodynamics === NBOMe-LAD is known to act as an agonist of the serotonin 5-HT2A receptor and to interact with other receptors, but shows dramatically reduced potency compared to LSD in vitro. At the serotonin 5-HT2A receptor, it had 37-fold lower affinity, 148-fold lower activational potency in terms of calcium release, and around half the maximal efficacy in terms of calcium release relative to LSD. On the other hand, NBOMe-LAD had only about 4-fold lower potency in terms of β-arrestin recruitment along with similar activational efficacy for this pathway relative to LSD. NBOMe-LAD produces the head-twitch response, a behavioral proxy of psychedelic effects, in rodents, but with greatly reduced potency and maximal efficacy relative to PRO-LAD and analogues. Its ED50Tooltip median effective dose for inducing the head-twitch response was 13-fold lower than that of PRO-LAD and its maximal effect was about one-third that of PRO-LAD. However, the most efficacious dose of NBOMe-LAD was 3.2 mg/kg whereas that of PRO-LAD was 1 mg/kg.
TAN-821 is an opioid drug which was originally reported as an agonist for the putative ε-opioid receptor, however this is now thought not to be a separate opioid receptor in its own right but is more likely a heteromer made from hybridisation of known opioid receptor subunits. It has subsequently been shown to have potent affinity for the kappa opioid receptor.
== Treatment == Treatment is focused on reducing stroke episodes and damage from a distending artery. Four treatment modalities have been reported in the treatment of vertebral artery dissection. The two main treatments involve medication: anticoagulation (using heparin and warfarin) and antiplatelet drugs (usually aspirin). More rarely, thrombolysis (medication that dissolves blood clots) may be administered, and occasionally obstruction may be treated with angioplasty and stenting. No randomized controlled trials have been performed to compare the different treatment modalities. Surgery is only used in exceptional cases.

It is a laboratory-made peptide that mimics growth hormone-releasing hormone. It prompts the pituitary gland to release growth hormone and has been studied mainly in adults with HIV-associated lipodystrophy.
Administered growth hormone supplies the hormone directly, while this peptide acts upstream by prompting the pituitary to release it. The indirect route preserves pulsatile secretion and some endogenous feedback, which changes the hormone and IGF-1 profile observed after treatment.
Reductions in visceral adipose tissue appear consistently in randomized trials of the approved population. Effects on peripheral fat, cardiovascular outcomes, and use outside that population are less well established.
No. It is a peptide that acts upstream of growth hormone release, while growth hormone is the hormone itself. The two differ in size, in receptor, and in how the body clears them.
